ABSTRACT

Acquired immune deficiency syndrome (AIDS) is a worldwide epidemic caused by human immunodeficiency virus (HIV) infection. Newer medicines for eliminating the viral reservoir and eradicating the virus are urgently needed. Attempts to locate relatively safe and non-toxic medications from natural resources are ongoing now. Natural-product-based antiviral candidates have been exploited to a limited extent. However, antiviral research is inadequate to counteract for the resistant patterns. Plant-derived bioactive compounds hold promise as powerful pharmacophore scaffolds, which have shown anti-HIV potential. This review focuses on a consideration of the virus, various possible HIV-controlling methods and the recent progress in alternative natural compounds with anti-HIV activity, with a particular emphasis on recent results from natural sources of anti-HIV agents. ABSTRACT Molecular methods have been responsible for a notable increase in the detection of Leishmaniinae infections in wild animals. Determining their infectiousness is of paramount importance in evaluating their epidemiological significance. One of the most efficient ways of determining infectiousness for vector borne diseases is xenodiagnosis with the appropriate vector. However, this is logistically very difficult to accomplish in the field, and an ideal solution is to find a molecular surrogate for xenodiagnosis. In this review we discuss different approaches to the problem by focusing on the infectiousness of Leishmania (Viannia) braziliensis in rodents under laboratory and field conditions. Comparisons with similar studies for other Leishmania species emphasizes that there are pivotal differences in the infectiousness and the importance of asymptomatic infections in different hosts. Potentially the most promising surrogate is the real time quantitative PCR (qPCR). However, its success depends on choosing a tissue that relates to the vector's feeding location and the parasite's tissue tropism. This requires detailed knowledge of the infection of each species in its wild hosts. We conclude that for L. (V.) braziliensis infections in wild rodents the tissue of choice for a molecular xenodiagnostic test, based on the qPCR is blood, providing that a significant number of samples must be examined.

ABSTRACT

BACKGROUND Elite controllers (EC) are human immunodeficiency virus (HIV)-positive individuals who can maintain low viral loads for extended periods without antiretroviral therapy due to multifactorial and individual characteristics. Most have a small HIV-1 reservoir composed of identical proviral sequences maintained by clonal expansion of infected CD4+ T cells. However, some have a more diverse peripheral blood mononuclear cell (PBMC)-associated HIV-1 reservoir with unique sequences. OBJECTIVES To understand the turnover dynamics of the PBMC-associated viral quasispecies in ECs with relatively diverse circulating proviral reservoirs. METHODS We performed single genome amplification of the env gene at three time points during six years in two EC with high intra-host HIV DNA diversity. FINDINGS Both EC displayed quite diverse PBMCs-associated viral quasispecies (mean env diversity = 1.9-4.1%) across all time-points comprising both identical proviruses that are probably clonally expanded and unique proviruses with evidence of ongoing evolution. HIV-1 env glycosylation pattern suggests that ancestral and evolving proviruses may display different phenotypes of resistance to broadly neutralising antibodies consistent with persistent immune pressure. Evolving viruses may progressively replace the ancestral ones or may remain as minor variants in the circulating proviral population. MAIN CONCLUSIONS These findings support that the high intra-host HIV-1 diversity of some EC resulted from long-term persistence of archival proviruses combined with the continuous reservoir's reseeding and low, but measurable, viral evolution despite undetectable viremia.

ABSTRACT

Ommaya reservoir implantation is generally used in the treatment of hydrocephalus and intraventricular drug administration. Ommaya reservoir implantation in the subarachnoid space of the spinal cord for the intrathecal drug administration has not been carried out in China, and only several reports can be retrieved from PubMed. About 60%-90% of untreated patients with spinal muscular atrophy type 2 (SMA2) who survive to adulthood often have complex scoliosis and joint deformities. Nusinersen is an effective drug for the treatment of SMA2. And the route of administration is intrathecal injection, which is difficult for patients with severe scoliosis. This article summarizes the process of Ommaya reservoir implantation and postoperative drug administration in a patient with complex scoliosis type SMA2, which provides a new method for clinical treatment of this disease.

Purpose: To investigate the impact of different?thickness scleral lenses (SLs) on corneal thickness, curvature, and fluid reservoir thickness in keratoconic eyes. Methods: Schiempflug imaging and AS?OCT was captured before and immediately following 6 h of SL wear. Different?thickness lenses were used while keeping the other parameters the same. The timing of the measurement for day 1 and day 2 was matched to allow for the control of the confounding influence of diurnal variation. Results: Immediately after 6 h of lens wear, no statistically significant difference (P > 0.05) was noted in corneal edema in any region and quadrants between thin? and thick?lens wearers. The calculated percentage of corneal edema was also within the range of overnight closed eye physiological swelling. Pentacam measured higher central corneal thickness compared to AS?OCT in both baselines and after 6 h of lens wear. The current investigation reported minimal but not statistically significant (P > 0.05) flattening in anterior and steepening in posterior curvature parameters in both thin and thick SLs. The mean reduction in the fluid reservoir thickness was 80.00 ± 3.99 and 79.36 ± 3.84 microns after 6 h of thin? and thick?lens wear, respectively, which was not statistically significant (P > 0.05). A statistically significant positive correlation (r = 0.67, P = 0.02) was found between lens thickness and change in anterior steep k with thick?lens wear. Conclusion: Central lens thickness of 200–400 ?m did not cause any significant change in corneal curvature and fluid reservoir thickness and did not induce clinically significant corneal edema after short?term SL wear.

Resumen Introducción: Los ríos tropicales se ven cada vez más afectados por la fragmentación y la regulación; y, en Colombia, se sabe que las represas ponen en peligro a los peces endémicos debido, entre otros, a la migración limitada y la disponibilidad reducida de redes alimenticias basadas en el detrito. Sin embargo, el conocimiento de la ictiofauna nativa afectada por represas en ríos altoandinos es aún incipiente. Objetivo: Evaluar los efectos de la represa del Neusa sobre la ictiofauna. Métodos: Comparamos dos secciones del río, una aguas arriba y otra aguas abajo de la represa con el Sistema Richter IHA, muestreamos tres transectos de 100 m de largo en cada sección, cada dos meses, entre 2017 y 2019. Los peces fueron devueltos al río después de efectuadas las mediciones corporales. Resultados: Recolectamos 729 individuos de cinco familias; los Trichomycterus bogotense eran más pequeños aguas abajo de la represa; Oncorhynchus mykiss fue más pequeño y menos abundante; y no hubo diferencias para Grundulus bogotensis y Eremophilus mutisii. Independientemente de los factores climáticos, O. mykiss y G. bogotensis fueron más abundantes aguas arriba, y E. mutisii y T. bogotense aguas abajo de la represa. Conclusión: Las cinco especies de peces diferían en cómo las poblaciones se diferencian aguas arriba y aguas abajo de la represa, lo que sugiere que algunas se benefician de la represa, mientras que otras se vuelven más pequeñas y menos abundantes.


Abstract Introduction: Tropical rivers are increasingly being affected by fragmentation and regulation; and, in Colombia, dams are known to endanger endemic fishes through, among others, limited migration and reduced availability of sediment-based feeding networks. However, knowledge of native ichthyofauna affected by dams in high Andean rivers is still incipient. Objective: To assess the effects of the Neusa dam on the ichthyofauna. Methods: We compared two rivers' sections, one above and one below the dam with the Richter IHA System, we sampled three 100 m long transects in each section, every two months, between 2017 and 2019. The fishes were returned to the river after body measurements. Results: We collected 729 individuals from five families; Trichomycterus bogotense were smaller under the dam; Oncorhynchus mykiss was smaller and less abundant; and there were no differences for Grundulus bogotensis and Eremophilus mutisii. Independently of climatic factors, O. mykiss and G. bogotensis were more abundant above the dam, and E. mutisii and T. bogotense under the dam. Conclusion: The five fish species differed in how the populations differed above and under the dam, suggesting that some are benefited by the dam, while others become smaller and less abundant.

Resumen Introducción: El embalse Cerrón Grande de 135 km2 es el humedal más grande de El Salvador, reconocido como "Humedal de Importancia Internacional" por la Convención Ramsar. Además, es uno de los ecosistemas más contaminados a nivel nacional, enfrentando serios problemas, entre ellos, las proliferaciones de cianobacterias. Objetivo: Evaluar las relaciones entre los factores ambientales y la distribución espacial y temporal de las cianobacterias en el embalse a lo largo de un año hidrológico. Métodos: Extrajimos agua con cianobacterias de seis sitios fijos, desde octubre 2018 hasta septiembre 2019. Resultados: Identificamos 16 géneros, el más abundante Microcystis sp. Además, Dolichospermum sp.; Microcistis sp. alcanzaron 1.5 x 106 células/ml en junio 2019, principalmente en el sector noroeste, que es poco profundo y cercano a las zonas de confluencia de afluentes altamente contaminados que llegan al embalse. El sector sureste más profundo tenía bajas concentraciones de estos organismos. Los factores ambientales mayormente relacionados con dicho comportamiento fueron: fósforo, la conductividad eléctrica y el valor de Secchi, como se esperaba de un cuerpo eutrófico. Conclusiones: La variación en la concentración celular de cianobacterias en este embalse está altamente influenciada por la lluvia, la mezcla de agua y el contenido de nutrientes.


Abstract Introduction: The 135 km2 Cerrón Grande reservoir is the largest wetland in El Salvador, recognized as "Wetland of International Importance" by the Ramsar Convention. It is also one of the most polluted ecosystems nationwide, facing serious problems, among them, a proliferation of cyanobacteria. Objective: To assess the relationships among environmental factors and the spatial and temporary distribution of cyanobacteria in the reservoir throughout a hydrological year. Methods: We extracted water with cyanobacteria from six fixed sites, from October 2018 through September 2019. Results: We identified 16 genera, the most abundant Microcystis sp. And Dolichospermum sp.; Microcystis sp. reached 1.5 x 106 cells/ml in June 2019, mainly in the north-west sector, which is shallow and close to the confluence zones where highly polluted tributaries reach the reservoir. The deeper south-east sector had low concentrations of these organisms. The environmental factors mostly related to this behavior were: phosphorus, electrical conductivity and Secchi value, as expected from a eutrophic body. Conclusions: Variation in the cellular concentration of cyanobacteria in this reservoir is highly influenced by rain, water mixing and nutrient content.

Resumen Las amebas de vida libre (AVL) son protozoos ubicuos con cuatro géneros patógenos para el ser humano: Acanthamoeba, Naegleria, Balamuthia, y Sappinia. Acanthamoeba puede actuar como reservorio de microorganismos (endosimbiontes), por lo cual, en medio hospitalario, implicaría un riesgo para la transmisión de bacterias, virus y hongos intranosocomiales. Se investigó la presencia de AVL, con énfasis en Acanthamoeba spp., en un hospital pediátrico de la provincia de Buenos Aires, Argentina. Se colectaron 22 muestras de lavamanos e incubadoras en salas de Neonatología y Terapia Intensiva, las que fueron cultivadas a 37 y 42 °C. Los aislados fueron identificados molecularmente. El 63,64% de las muestras presentaron Acanthamoeba spp. Esta investigación representa el primer estudio realizado en la Argentina sobre la detección de Acanthamoeba spp. en salas cerradas de un hospital. Su presencia es una señal de alarma y resulta un blanco útil para investigar posibles reservorios de microorganismos patógenos en ambientes hospitalarios.


Abstract Free-living amoebae (FLA) are ubiquitous protozoa with four pathogenic genera for humans: Acanthamoeba, Naegleria, Balamuthia, and Sappinia. Acanthamoeba can act as a reservoir of microorganisms (endosymbionts), for which reason, in a hospital environment, it would imply a risk for transmission of nosocomial bacteria, viruses and fungi. The presence of AVL, with emphasis on Acanthamoeba spp., was investigated in a pediatric hospital. Twenty-two samples were collected from sinks and incubators in Neonatology and Intensive Care rooms, which were cultured at 37 and 42 °C. The isolates found were molecularly identified. A total of 63.64% of the samples presented Acanthamoeba spp. This research represents the first study in Argentina on the detection of Acanthamoeba spp. in closed rooms of a hospital. Its presence is an alarm signal, and it is a useful target to investigate possible reservoirs of pathogenic microorganisms in hospital environments.

Vinte e duas amostras foram coletadas em lavatórios e incubadoras em Salas de Neonatologia e Cuidados Intensivos, cultivadas a 37 e 42 °C. Os isolados foram identificadas molecularmente. Foram encontradas Acanthamoeba spp. em 63,64% das amostras. Esta investigação representa o primeiro estudo realizado na Argentina sobre a detecção de Acanthamoeba spp. em salas fechadas de um hospital. A sua presença é um sinal de alarme e um alvo para investigar possíveis reservatórios de microrganismos patogênicos em ambientes hospitalares.

ABSTRACT

Present study has been undertaken in order to assemble knowledge regarding physicochemical parameters analysis from June 2021 – May 2022 in relation to the abundance, composition and seasonal variation in the phytoplankton of Johila reservoir Lalpur Podki, Amarkantak (M.P.). It is interiorly located at Latitude 22046'20, Longitude, 81044'39'' in Lalpur and no study regarding hydrobiology in general and phytoplankton abundance in particular has yet not been conducted. Johila reservoir has its origin from Jwaleshwar temple kund of Amarkantak, hill located in Makel velly. It starts flowing opposite site to the Narmada River after 15 Kilometre and finally joins the river Sone at Dasrathghat of Umaria District of Madhya Pradesh. Total area of Johila reservoir is 86 hectare (216 acers). Since no study yet has been done on phytoplankton production in relation to its physico-chemical characteristic hence present study has been undertaken. Phytoplankton population were dominated by Chlorophyceae, Myxophyceae and Bacillariophyceae in decreasing order. Chlorophyceae members revealed dominance over Myxophyceae and Bacillariophyceae.

Abstract Hantavirus Pulmonary Syndrome (HPS) is an emerging infectious disease of the Americas. Eight native rodent species have been identified as HPS virus reservoirs in Argentina. The aim of this work was to detect the orthohantavirus genotypes present in a rodent commu-nity that inhabits a zone where a fatal HPS case occurred within an endemic locality of Central Argentina. We captured 27 rodents with a trapping effort of 723 trap nights. We detected 14.3% of infected Akodon azarae with the Pergamino genotype. This result expands the known distribution of this orthohantavirus. Although the Pergamino genotype has not been associated with human cases, the information about its distribution is relevant for risk assessment against potential changes in the virus infectivity.


Resumen El síndrome pulmonar por hantavirus (SPH) es una enfermedad infecciosa emergente en América. Ocho especies de roedores nativos han sido identificadas como reservorios del virus causante del SPH en la Argentina. El objetivo de este trabajo fue detectar los genotipos de orthohantavirus presentes en una comunidad de roedores que habita en una zona donde ocurrió un caso fatal de SPH, en una localidad endémica de Argentina central. Se capturaron 27 individuos con un esfuerzo de 723 trampas-noche. Se detectó un 14,3% de Akodon azarae infectados con el genotipo pergamino. Este resultado amplía el conocido rango de distribución de este orthohantavirus. A pesar de que el genotipo pergamino no ha sido asociado con casos humanos hasta el momento, la información sobre su distribución es relevante para analizar el riesgo ante un potencial cambio en la infectividad del virus.

Porcine circovirus 2 (PCV2) has a considerable economic impact on the pork industry worldwide for more than two decades. In 2016, a new circovirus, porcine circovirus 3 (PCV3), was described; since then, it has been reported to be associated with diseased or even in clinically healthy swine in several countries. Considering the importance of wild boars as reservoirs of swine pathogens and the extensive distribution of these animals in Rio Grande do Sul and throughout the national territory, we searched for PCV2 and PCV3 in twenty-six wild boars coupled with necropsy and histologic examination of the sampled animals. Using PCR, 182 tissue samples were analyzed, including the heart, kidneys, liver, lung, lymph nodes, spleen, and tonsils. PCV2 and PCV3 were detected in 57.7% (15/26) and 15.4% (4/26) of wild boars, respectively. Furthermore, co-infection with PCV2 and PCV3 was detected in one of these animals, with PCV2 or PCV3 DNA detection in multiple organs. Histological examination showed mild to moderate and multifocal lymphoplasmacytic interstitial nephritis distributed randomly throughout the renal cortex, apparently unrelated to PCV2 or PCV3 detection. The wild boar population in Brazil is extensive, indicating the presence of a larger number of swine pathogen hosts. In the present study, more than half of the wild boars harbored PCV2; and although less frequently, PCV3 was also detected. Therefore, free-living wild boars can serve as reservoirs of swine circoviruses in southern Brazil.


O circovírus suíno 2 (PCV2) tem causado impacto econômico na indústria suína em todo o mundo por mais de duas décadas. Em 2016, um novo circovírus foi descrito - circovírus suíno 3 (PCV3) - e desde então tem sido relatado em vários países associado a doenças ou mesmo suínos saudáveis. Diante da importância dos javalis como reservatórios de patógenos suínos, e da ampla distribuição desses animais no Rio Grande do Sul e em todo o território nacional, foi realizada pesquisa de PCV2 e PCV3 em vinte e seis javalis (10 fêmeas e 16 machos). Necropsia e exame histológico foram realizados. Utilizando PCR, foram analisadas 182 amostras de tecidos incluindo: coração, rins, fígado, pulmão, linfonodos, baço e tonsila. PCV2 e PCV3 foram detectados por PCR em 57,7% (15/26) e 15,4% (4/26) dos javalis, respectivamente. Um destes animais estava co-infectado por PCV2 e PCV3. O DNA do PCV2 ou PCV3 foi detectado em multiplos órgãos. No exame histológico foi observada nefrite intersticial linfoplasmocitária multifocal leve a moderada, distribuída aleatoriamente pelo córtex renal, aparentemente sem relação com a detecção de DNA viral. A população de javalis no Brasil é extensa, resultando em maior número de hospedeiros para patógenos de suínos. No presente estudo, mais da metade dos javalis capturados abrigavam PCV2 e, embora menos frequente, PCV3 também foi detectado. Os javalis de vida livre podem servir como reservatórios de circovírus suínos no sul do Brasil.

ABSTRACT

Chapare hemorrhagic fever is an emerging zoonotic disease caused by a Mammarenavirus, of the Arenaviridae family, called Chapare virus. Its name refers to the region in which it first appeared. This virus is transmitted through direct contact with the excreta of wild rodents that interact with humans by invading homes close to a rural area and when humans enter the jungle for various reasons such as agriculture and tourism, among others. This virus has been classified by the World Health Organization as biosafety level IV, where pathogens with the highest risk for humans are included and for which there are no vaccines available. It produces a febrile and hemorrhagic picture that leads to death two weeks after the onset of symptoms. There is no vaccine or standardized treatment to treat this disease. The bio ecological aspects of the reservoir are not known, nor are the factors that could be related to the emergency in the area. An investigation should be carried out focused mainly on knowing the circulation of the Chapare virus in the Bolivian tropics, knowing the bioecological characteristics of the reservoir to produce and contribute with reliable information for the design of future surveillance and control strategies with community participation, as well as to strengthen laboratory diagnostic capacities in health establishments in the municipality of Villa Tunari.


La fiebre hemorrágica Chapare es una enfermedad zoonótica emergente causada por un Mammarenavirus de la familia Arenaviridae, llamado virus Chapare; su nombre se debe a la región en la cual apareció por primera vez. Este virus es transmitido por medio del contacto directo con las excretas de los roedores silvestres que interactúan con el ser humano, al invadir las viviendas que se encuentran cercanas al área silvestre y cuando el hombre incursiona a la selva por diversas razones, como las actividades de agricultura o turismo, entre otras. Este virus ha sido catalogado por la Organización Mundial de la Salud como nivel de bioseguridad IV, en el cual se ubican los patógenos de mayor riesgo para el ser humano y para los cuales no existen vacunas disponibles. Produce un cuadro febril y hemorrágico que lleva al fallecimiento a las dos semanas de haber iniciado los síntomas. No existe vacuna ni tratamiento estandarizado para tratar esta enfermedad, no se conocen los aspectos bioecológicos del reservorio y tampoco los factores que podrían estar relacionados con la emergencia en la zona. Se deberá realizar una investigación enfocada principalmente en conocer la circulación del virus Chapare en la región del trópico boliviano, conocer las características bioecológicas del reservorio con el propósito de producir y aportar con información fehaciente para el diseño de futuras estrategias de vigilancia y control con participación comunitaria, así como fortalecer las capacidades de diagnóstico de laboratorio en los establecimientos de salud del municipio de Villa Tunari.

Background: One of the main health problems in West Africa remains upsurge of emerging pathogens. Ebola virus disease outbreak occurred in 2014 in Liberia, Guinea and Sierra Leone, Monkeypox virus in Nigeria in 2017 and most recently Lassa virus in Nigeria, Togo and Benin in 2018. These pathogens have animal reservoirs as vectors for transmission. Proper investigation of the pathogens in their rodent vectors could help reduce and manage their emergence and spread. Methodology: This study was conducted with an approval from the Côte d'Ivoire Bioethics Community. Small mammal trappings were carried out in 9 sites within three zones namely, peri-urban, peri-rural and protected areas. Liver, lung and kidney tissues from trapped small mammals were sampled in accordance with the recommended conditions of biosafety and bioethics. The organs were transported in liquid nitrogen to the laboratory. Molecular tests were used to detect pathogens. Orthopoxviruses and Monkeypox virus were detected in the organs by PCR using consensus primers targeting the virus surface membrane haemagglutinin (HA) genes, while Leptospira species were detected by PCR using primers targeting the rrs and lfb1 genes. Results: Out of 4930 night-traps, 256 (5.19%) small mammals were trapped including Crocidura, Rattus, Lophuromys, Praomys, Mus and Mastomys. Leptospira species were detected in 6 genera from 7 study sites and the infected small mammals accounted for 13.3%. Leptospira sp was detected mainly in the rodent vector genera Rattus (32.3%), Lophuromys (29.0%), and Praomys (16.1%). Three species of Leptospira were detected and Leptospira interrogans was the most common frequent species (74.2%). Monkeypox virus was not detected from studied small mammals. Conclusion: The initial data from our investigation indicates the presence of Leptospira sp in rodent vectors, Rattus, Lophuromys and Praomys, which are the potential small mammalian reservoirs of this pathogen in Cote d'Ivoire.

Resumen Introducción: Enterobacterales productores de carbapenemasas (EPC) son una importante causa de infecciones asociadas a la atención en salud (IAAS). El principal reservorio de EPC lo constituyen pacientes infectados y colonizados, sin embargo, también se han identificado reservorios ambientales. Objetivo: Detectar la presencia de EPC en los sifones de lavamanos de la unidad de cuidados críticos de pacientes quemados adultos (UPC QMD) y unidad de cuidados críticos de pacientes pediátricos crónicos (UCEP). Método: Se recolectaron cuatro muestras de sifones de los lavamanos ubicados en el interior de las unidades de pacientes en UCEP y 10 de UPC QMD. A las muestras se les realizó estudio fenotípico y molecular para detección de carbapenemasas en el Instituto de Salud Pública de Chile. Resultados: En los sifones estudiados de UCEP no se aislaron cepas de EPC. En UPC QMD, 50% de los sifones estudiados se aislaron cepas de EPC. Conclusiones: En UPC QMD se objetivó la presencia de EPC en una alta proporción de los sifones de lavamanos testeados, lo que demuestra un reservorio ambiental de bacterias multi-resistentes.


Abstract Introduction: Carbapenemase-producing Enterobacterales (CPE) are an important cause of health care associated infections (HAI). The main reservoir is constituted by infected and colonized patients; however, environmental reservoirs have also been identified. Objective: To detect the presence of CPE in the sink traps of the critical care unit for adult burn patients (UPC QMD) and the critical care unit for chronic pediatric patients (UCEP). Material and Method: Four samples of trap were collected from the sinks located inside the patient units at PICU and 10 at UPC QMD. The samples underwent a phenotypic and molecular study for the detection of carbapenemases at the Institute of Public Health of Chile. Results: In the UCEP no EPC strains were isolated. In UPC QMD, CPE was detected in 50% of the traps. Conclusions: In UPC QMD, the presence of CPE was observed in a high proportion of the tested sinks traps, which shows an environmental reservoir of multi-resistant bacteria.

ABSTRACT

ABSTRACT Zika virus (ZIKV) is an enveloped, single-stranded RNA arbovirus belonging to the genus Flavivirus. It was first isolated from a sentinel monkey in Uganda in 1947. More recently, ZIKV has undergone rapid geographic expansion and has been responsible for outbreaks in Southeast Asia, the Pacific Islands, and America. In this review, we have highlighted the influence of viral genetic variants on ZIKV pathogenesis. Two major ZIKV genotypes (African and Asian) have been identified. The Asian genotype is subdivided into Southwest Asia, Pacific Island, and American strains, and is responsible for most outbreaks. Non-synonymous mutations in ZIKV proteins C, prM, E, NS1, NS2A, NS2B, NS3, and NS4B were found to have a higher prevalence and association with virulent strains of the Asian genotype. Consequently, the Asian genotype appears to have acquired higher cellular permissiveness, tissue persistence, and viral tropism in human neural cells. Therefore, mutations in specific coding regions of the Asian genotype may enhance ZIKV infectivity. Considering that mutations in the genomes of emerging viruses may lead to new virulent variants in humans, there is a potential for the re-emergence of new ZIKV cases in the future.

Objective:To analyze the correlation between human immunodeficiency virus (HIV)-1 reservoir and poor immune reconstitution of HIV/acquired immunodeficiency syndrome (AIDS) patients, and to investigate the influence of HIV-1 reservoir on the immune reconstitution.Methods:Cross-sectional survey was conducted to measure HIV-1 RNA and T lymphocyte subsets from 219 patients with HIV/AIDS who had been treated with anti-retroviral therapy (ART) for more than two years with HIV RNA lower than the limit of detection. Among them, there are 195 patients from the Sixth People′s Hospital of Zhengzhou, 12 patients from Shangqiu Municipal Hospital and 12 patients from Zhoukou Infectious Diseases Hospital. Peripheral blood mononuclear cells (PBMC) were collected and HIV-1 DNA was detected. The measurement data of normal distribution were analyzed by two independent sample t-test. The measurement data of skewness distribution were analyzed by rank sum test. Spearman′s rank correlation was used for correlation analysis. Receiver operating characteristic curve (ROC) was used to predict the predictive value of occurrence of poor immune reconstitution AIDS patients. Results:There were 121 patients with poor immune reconstitution and 98 patients with healthy immune reconstitution. HIV-1 DNA was (2.50±0.52) copies/1×10 6 PBMC in the group with poor immune reconstitution, which was significantly higher than the healthy immune reconstitution group ((2.11±0.66) copies/1×10 6 PBMC, t=4.78, P<0.001). The CD4 + T lymphocyte counts in the group with poor immune reconstitution was 192(139, 227)/μL, which was lower than that in the healthy immune reconstitution group (573(457, 730)/μL). The difference was statistically significant ( Z=12.68, P<0.001). HIV-1 DNA was reversely correlated with CD4 + T lymphocyte counts and CD4 + /CD8 + T lymphocyte ratio (after adjusting the influence of age and ART time, r=-0.277 and -0.316, respectively, both P<0.001). The area of ROC curve for HIV-1 DNA to predict poor immune reconstitution was 0.679(95% confidence interval ( CI) 0.604 to 0.750). The HIV-1 DNA threshold value was 100 copies/1×10 6 PBMC with the sensitivity of 90.13% and specificity of 42.91%. The area of ROC curve of CD4 + /CD8 + T lymphocyte ratio to predict poor immune reconstitution was 0.905 (95% CI 0.863 to 0.942). The threshold value of CD4 + /CD8 + T lymphocyte ratio was 0.536 with the sensitivity of 77.68% and specificity of 89.84%. Conclusions:There is correlation between HIV-1 DNA and poor immune reconstitution in HIV/AIDS patients. The value of HIV-1 DNA higher than 100 copies/1×10 6 PBMC and CD4 + /CD8 + T lymphocyte ratio lower than 0.536 could be used as predictor of poor immune reconstitution.

ABSTRACT The understanding of the relationships between the planktonic communities in a reservoir allows us to infer possible changes in the redistribution of matter and energy flows in these systems. This work proposes a dynamic model for the trophic network of the Riogrande II tropical reservoir, which integrates the planktonic trophic chains of detritus and grazing, limiting the prey-predator interactions by introducing the prey meeting factor (pmf). We built a dynamic model of mass balance supported by an extensive bibliographic search. The limitations of consumers and resources were represented simultaneously by means of the pmf. The data used to validate the model were compiled from previous investigations carried out in this reservoir from 2010 to 2013. The values of pmf that we found in each simulation suggest that the top predator can access its main prey in certain concentrations of total phosphorus, with a probability of encounter ranging from 9.3 % to 17.7 %. Our simulations indicate that most of the primary production is poorly used by the primary consumers in the photic zone, however, it enters in the flows of the detrital chain and supports the production of zooplankton almost entirely. According to this finding, the biomass densities obtained in the previous studies can be better explained by the causal relationships assumed in this model.


RESUMEN Entender las relaciones entre las comunidades planctónicas en un embalse nos permite inferir posibles cambios en la redistribución de los flujos de materia y energía en este sistema. Este trabajo propone un modelo dinámico para representar la red trófica del embalse tropical Riogrande II, donde se integran las cadenas tróficas de pastoreo y detritus y se limitan las interacciones entre predadores, presas y recursos al introducir un factor limitante de encuentro con la presa (pmf). El modelo dinámico se enfoca en el balance de masas sustentado en una amplia búsqueda bibliográfica. Los datos usados para validar el modelo se colectaron de datos previamente reportados para el embalse durante los años 2010 y 2013. Los valores de pmf obtenidos en cada simulación, sugieren que el predador dominante puede acceder a su presa principal a ciertas concentraciones de fósforo total, con una probabilidad de encuentro que va desde 9,3 % hasta 17,7 %. Nuestros resultados indican que la mayor parte de la producción primaria es poco aprovechada por los consumidores en la zona fótica, sin embargo, ingresa en el flujo de la cadena detrítica de manera que soporta la producción de zooplancton casi por completo. Las relaciones causales asumidas en este modelo explican en gran medida las densidades de biomasa reportadas en estudios previos.

Introducción: Los craneofaringiomas son tumores benignos frecuentes en pediatría. La cirugía resectiva solía ser el tratamiento de elección. Sin embargo, en los últimos años se encuentra muy cuestionada debido a su elevada morbilidad. El objetivo primario de este trabajo es presentar los resultados clínicos y quirúrgicos de la cirugía resectiva de craneofaringiomas por vía transcraneal en nuestra institución. El objetivo secundario es presentar una serie de casos en los que se realizó un tratamiento quirúrgico conservador utilizando un reservorio de Ommaya para tratamiento del componente quístico. Métodos: Estudio descriptivo de una serie de casos que compara evaluaciones clínicas e imagenológicas pre y postoperatoria en pacientes sometidos a cirugía resectiva de craneofaringioma. Además, se presenta una serie de casos de pacientes con craneofaringiomas predominantemente quísticos tratados con colocación de catéter de Ommaya intraquístico e Interferón. Resultados: Se analizaron 33 pacientes con craneofaringioma sometidos a resección quirúrgica. Al año de la cirugía, encontramos que el 51.52% de los casos (17 pacientes) se mantuvo sin crecimiento de la lesión y que el 48.48% (16 pacientes) presentó aumento o recidiva. En el postoperatorio todas las evaluaciones clínicas empeoraron. Fueron estadísticamente significativas la aparición de diabetes insípida (18.18% vs. 69.70%; p=0.04), obesidad (12.12% vs. 36.36%; p=0.005) y talla baja (33.33% vs. 57.58%; p=0.0006). En los tratados con Ommaya e Interferón, el 28.57% (2 casos) presentó reducción o estabilidad en el quiste y el 71.43% (5 casos) aumentó el tamaño, aunque presentó mejores resultados respecto a la restricción de la talla y obesidad. Conclusión: La exéresis quirúrgica de los craneofaringiomas genera una alta tasa de comorbilidades sumado a un porcentaje elevado de recidiva. El manejo conservador tratando el componente quístico en los casos en los que sea posible podría ser una opción viable. El tratamiento de los craneofaringiomas de la infancia debe abordarse de forma multidisciplinaria intentando preservar el bienestar y la calidad de vida de los pacientes


Introduction: Craniopharyngioma is a common benign tumor in children. Although surgery has traditionally been the treatment of choice, it has been controversial in recent years due to the associated high morbidity. The main aim of this paper is to present the clinical and surgical outcomes seen after craniopharyngioma resection in our institution. The secondary objective is to present a case series of patients treated with an Ommaya reservoir (OR) and interferon. Methods: This was a descriptive study comparing the pre and post-surgical clinical and image assessment in patients undergoing craniopharyngioma resection. Also, a case series including patients with craniopharyngioma managed with an Ommaya reservoir (OR) and Interpheron was included. Results: Thirty-three patients with craniopharyngioma undergoing surgical resection were assessed. We found no tumor growth in 51.52% (17) of the cases, and either tumor growth or recurrence in 48.48% of the cases at one year. Clinical worsening was observed in the post-operative period; the presence of diabetes insipidus (18.18% vs. 69.70%; p=0.04), obesity (12.12% vs. 36.36%; p=0.005) and short stature (33.33% vs. 57.58%; p=0.0006) were statistically significant. Of the patients treated with an OR and interferon, 28.57% (2) presented tumor cyst regression or stability, and 71.43% exhibited tumor cyst growth. Conclusion: The surgical resection of craniopharyngiomas is associated with a high recurrence rate and usually high mortality. A multidisciplinary management of craniopharyngiomas in childhood is advisable in order to preserve the wellbeing and quality of life of patients

The introduction of non-native predators is a matter of great concern, but their impacts on ecosystem functions remain poorly understood. We investigated how changes in fish diversity following the invasion of Cichla kelberi affected ecosystem functions generated by fish populations. Fish assemblages were sampled in macrophyte patches in a Neotropical impoundment over a 5-year period, before and after the introduction of the predator. We assigned seven ecosystem functions (26 trait-states) to each fish species, and examined how these functions behaved after the invasion. We collected 577 fish belonging to 25 species. Species richness, fish biomass and main species declined significantly over periods. The biomass of ecosystem functions changed significantly over time, and most trait-states declined. Few trait-states were lost, but all functions had at least one trait-state reduced by more than 85%. A null model analysis showed that changes in functions were not driven by species identities, while species richness correlated positively with total biomass and with most functions, suggesting that the loss of taxa and biomass drove observed changes in ecosystem functions. Our study provided evidence that community disassembly associated with the invasion of C. kelberi translated to the decline of several ecosystem functions, affecting energy mobilization and transference.(AU)


A introdução de predadores não-nativos tem gerado grande preocupação, mas seus impactos sobre a geração de funções ecossistêmicas permanecem pouco investigados. O presente estudo investigou como mudanças na diversidade de peixes, associadas com a invasão de Cichla kelberi, afetaram funções ecossistêmicas geradas por populações de peixes. As assembleias de peixes foram amostradas em bancos de macrófitas em um grande reservatório Neotropical durante um período de cinco anos, antes e depois da introdução do predador. Um conjunto de funções ecossistêmicas (26 trait-states) foi atribuído a cada espécie de peixe, e examinamos como as funções se comportaram depois da invasão. Foram coletados 577 peixes pertencentes a 25 espécies. Registramos declínio significativo da riqueza de espécies, biomassa total e biomassa das principais espécies ao longo dos períodos. A biomassa das funções ecossistêmicas mudou significativamente ao longo do tempo, e a maioria dos trait-states declinou. Poucos trait-states foram perdidos, mas todas as funções tiveram pelo menos um trait-state reduzido em mais de 85%. Uma análise de modelos nulos mostrou que as mudanças nas funções não foram impulsionadas pela identidade das espécies, enquanto que a riqueza de espécies correlacionou positivamente com a biomassa total e com a maioria das funções, sugerindo que a perda de espécies e biomassa impulsionou as mudanças observadas nas funções ecossistêmicas. Nosso estudo apresenta evidências de que a desestruturação da comunidade, associada com a invasão de C. kelberi, se traduziu no declínio de várias funções ecossistêmicas, afetando a mobilização e transferência de energia.(AU)

ABSTRACT

We investigated changes in the species richness, abundance, and composition of the zooplankton community in response to the formation of a small reservoir in the Caveiras River, southern Brazil. Zooplankton were collected using a motor-pump and aplankton net (68 μm mesh), with 600 L of water filtered per sample. Sampling occurred during the pre-(April, August, and December 2011) and post-impoundment (July and October 2013, and January 2014) phases of the Caveiras River. We identified 86 taxa in this study, and rotifers were the predominant group. The species richness and abundance of the zooplankton increased after the filling of the reservoir. Furthermore, the zooplankton community showed a clear change in the species composition between the phases before and after the formation of the reservoir, with the emergence of typical planktonic species. Changes in the structure of the zooplankton community were related to changes in limnological characteristics due to the impoundment of the river, mainlyin the availability of food and in the concentration of nutrients.
